چکیده
Improved prognosis for triple-negative breast cancer (TNBC) has plateaued and the development of novel therapeutic strategies is required. This study aimed to explore anti-tumor effect combined eribulin HDAC inhibitor (vorinostat: VOR, pan-HDAC ricolinostat: RICO, selective HDAC6 inhibitor) treatment TNBC. The in combination with an was tested three TNBC cell lines (MDA-MB-231, Hs578T, MDA-MB-157) their eribulin-resistant derivatives. expression acetylated ?-tubulin analyzed by Western blotting cells immunohistochemical analyses clinical specimens obtained from patients who were treated eribulin. simultaneous administration low concentrations (0.2 ?M) VOR or RICO enhanced MDA-MB-231 Hs578T but not MDA-MB-157 cells. Meanwhile, pretreatment 5 ?M sensitivity all lines. Low concentration increased In contrast, whereas cells, did not. Eribulin These phenomena also observed Immunohistochemical revealed that after inhibition enhances through acetylation ?-tubulin. therapy could represent a strategy

Synergistic effect of eribulin and CDK inhibition for the treatment of triple negative breast cancer
Activation of CDK2 in triple negative breast cancer (TNBC) can contribute to non-canonical phosphorylation of a TGFβ signaling component, Smad3, promoting cell proliferation and migration. Inhibition of CDK2 was shown to decrease breast cancer oncogenesis. Eribulin chemotherapy was used effectively in the treatment of TNBC. To this end, we tested therapeutic efficacy of a novel CDK2/9 inhibitor...
متن کاملCytotoxicity and anti-tumor effects of new ruthenium complexes on triple negative breast cancer cells
Triple-negative breast cancer (TNBC) is a highly aggressive breast cancer subtype. The high rate of metastasis associated to the fact that these cells frequently display multidrug resistance, make the treatment of metastatic disease difficult. Development of antitumor metal-based drugs was started with the discovery of cisplatin, however, the severe side effects represent a limitation for its c...

Background: Triple-negative breast cancer (TNBC) is treated with highly aggressive non-targeted chemotherapies. Safer and more effective therapeutic approaches than those currently in use are needed. Natural pomegranate peel extract (PPE) has recently been found to inhibit breast cancer progression; however, its mechanisms of action remain unclear. We hypothesized that transcriptional chan...
متن کاملClinicopathological Features of Non-metastatic Triple Negative Breast Cancer
Background: Triple negative breast cancer (TNBC) is reported to be associated with a high risk of recurrence, poor overall survival (OS), and disease-free survival (DFS) rates. This study evaluated the clincopathological features and survival of non-metastatic TNBC women in the capital of Iran compared with other areas of the world. Methods: In a retrospective study, 119 women with TNBC based ...
متن کاملMicrotubule acetylation through HDAC6 inhibition results in increased transfection efficiency.
The success of viral and nonviral gene delivery relies on the ability of DNA-based vectors to traverse the cytoplasm and reach the nucleus. We, as well as other researchers, have shown that plasmids utilize the microtubule network and its associated motor proteins to traffic toward the nucleus. While disruption of microtubules with nocodazole was shown to greatly inhibit cytoplasmic plasmid tra...
